titleOfInvention Method of determining antibodies to virus of manъs immunodeficiency in blood
abstract The invention relates to the field of medicine, specifically to the immunodiagnostics of acquired immunodeficiency syndrome, and can be used to determine the presence of antibodies to proteins of the HIV virus in the blood. The goal is to increase the sensitivity and simplify the method. In the proposed method of detecting antibodies to HIV proteins, SDS electrophoresis in a gradient 4-20% acrylamide gel is used, virus-containing material - extracts of virus-producing cells of the lymphoblastoid line NTA-4. Separated in acrylamide gel to individual protein bands, HIV antigens are transferred by electromigration to a nitrocellulose filter, viral antigens are identified and nitrocellulose filters bands corresponding to p24, p55, other 41, dr 120 proteins are cut and fragments of these bands are mounted with nitrocellulose glue on a special paper plate , forming a diagnostic strip. The active part of the Diagnostic strip, which consists of 4 fragments of nitrocellulose filters, each of which carries one HIV protein on its surface: p24, p55, others 41, others 120, is immersed in the analyzed serum, diluted 1:50 with saline. After incubation, a diagnostic strip is developed using an anti-species conjugate, similarly to the immunological development of the prototype method - Western blot. In order to increase the reliability of the assay results in the ELISA, two marker enzymes, β-lactamase from BACILLUS LICHENIFORMIS and horseradish peroxidase, are used.
