abstract | The invention relates to biochemistry, in particular, to clinical biochemistry, and can be used to diagnose diseases. The aim of the invention is to improve the accuracy of the method. For this purpose, 0.5 ml of 27.6-32.2 mmol / l solution of nicotine-amide nucleotide (NAD) and 2.0 ml of 0.4-0.5 mol / l bicarbonate buffer PH 10.0-10 are taken in the test sample. , 5, containing 63 mmol / l glutamate. In the control sample add 0.5 ml of NAD of the same concentration and 2.0 ml of 0.4-0.5 mol / l bicarbonate buffer without glutamate. Then, 0.1 ml of biological fluid is added to the test and control samples, followed by incubation at room temperature for 10-15 minutes and measuring the optical density at 365 nm. The activity (D) is determined by the increment of the reduced nicotinamiddine nucleide (NADH). Per unit of activity, the amount of enzyme is taken, which in 1 minute increases the content of NADH in the sample by 1 μmol. The result is calculated on 1 l serum and expressed in ED. The method is simple to perform, does not require large volumes of the studied biological fluid, 2 times more accurate than the known. |
