abstract | The invention relates to peptides, in particular to the preparation of N-acetylmuramoyl-L-alanyl-D-isoglutamine β-6-aminohexylglucoside hydrochloride, which is used to prepare conjugates with synthetic and natural polymeric carriers. The goal is to increase the yield of the target product. The preparation is carried out by use in the synthesis of oxazoline as a starting material of 1,6-hexanediol obtained paratsetat β-6-N-acetylglucosamine gidroksilglikozid mesylated mesyl chloride in pyridine followed by conversion of the mesylate to azide benzilideniruyut benzaldehyde dimethylacetal without a solvent, is condensed with protected acid trifluoroacetate benzyl ester of L-ananyl-D-isoglutamine with N-oxysuccinimide and N, Nb-dicyclohexylcarbodiimide, benzylidene protection is removed by acid hydrolysis and hydrogenated over kisyu palladium in the presence of chloroform. The method allows to isolate the desired product as hydrochloride. The total yield of 22% (vs. 7% in the known method). |
