titleOfInvention | Method of predicting the development of angionephropathy in diabetes mellitus of children |
abstract | The invention relates to medicine, in particular, to endocrinology, and can be used to predict angiophrotic complications in diabetes mellitus in children. The aim of the invention is to improve the accuracy of the method. For this purpose, the protein content in the patient's daily urine is determined, and fibronectin in the blood. In a urine sample of 1.25 ml, 3.75 ml of 3% sulfasalicylic acid is added, and a control with 1.25 ml of distilled water is placed in parallel. After exposure, the protein content is determined by photoelectric colorimeter readings (α = 590, light orange) using a calibration curve. To obtain plasma, blood is mixed with 3.5% trisubstituted sodium citrate in a 9: 1 ratio. Fill the glass plate with a solution of agarose, add a 3% solution of fibronectin antisera. When the agar dries out, a 2.5 mm diameter hole is drilled with a punch and a 6 µl plasma is applied using a Hamilton syringe. At the same time, they will monitor plasma samples in dilutions of 1: 2, 1: 4, 1: 8. The concentration of fibronectin in the sample is 300 µg / ml. Electrophoresis is carried out on an agar gel containing monospecific antiserum. The solutions are added to the wells and stained after 12 hours. The identification of fibronectin is carried out by the formation of a precipitation peak in the form of a rocket. After that, the ratio of the protein content in the urine to the content of fibronectin in the blood is calculated and, at a value of 0.2-0.8 standard units, predict the development of angionephropathy for 6-12 months in diabetes mellitus in children. The prediction accuracy is 100%. |
