titleOfInvention Method of quantitative determination of lignin in plants
abstract This invention relates to the analytical chemistry of natural compounds. The purpose of the invention is to increase the reliability of the analysis. Lignin is extracted from plant material by alkaline hydrolysis with 1-2% NaOH solution at 150-165 ° C for 1-3 hours. Extracted lignin is separated from interfering impurities in the extract by filtering the extract, acidifying it with 3-5% HCl prior to sedimentation of lignin, separation of lignin on a chromatographic column with diatomite deactivated with hexamethyl disilazone, and subsequent depletion of it from the column with dimethyl sulfoxide. In the resulting solution, the lignin content was determined by spectrophotometry in the UV spectrum. The method makes it possible to obtain reliable data on the content of lignin in all plant species with a sensitivity of 0.001% and a relative standard deviation not exceeding 0.05. 3 dw., 2 tabl.
