titleOfInvention Strain of hybrid cultivatable cells of mus musculus mice, used for producing monoclonal antibodies to human urokinase
abstract This invention relates to biotechnology and can be used to isolate, purify, and determine urokinase in biological fluids. The aim of the invention is to obtain a hybrid strain of cultured mouse MUS musculus cells producing monoclonal antibodies (MA) to human urokinase, which have an increased antigen binding constant and interact with two molecular forms of the enzyme. The UIG strain is obtained by hybridization of splenocytes of Balb / c mice immunized with a urokinase preparation with X63-Ag8.653 myeloma cells. It is stored in the Specialized Collection of Intertwined Somatic Cells of the Vertebrates of the Institute of Cytology, Academy of Sciences of the USSR under the number BCKK (n) 35D and is characterized by the following signs. Cultivation medium - RPMI-1640 medium with 10% zmbrional calf and 10% horse serum, sowing dose of 10 cells. in 1 ml, the multiplicity of sieving 1:10 every 3-4 days, culture suspension. The strain is transported as ascites in Balb / c mice at least 3 times. The content of MA in the culture liquid on day 4 of cultivation is 20-30 µg / ml, in mature ascites - 2-3 mg / ml. ML belongs to class 1 G, t (K-5, have a constant of binding to antigen, inhibit the activity of urokinase, interact with its two molecular forms. MA are used to determine and purify urokinase.
