titleOfInvention Stock culture of hybrid cultivated cells of musculus mouse used for producing monoclonal antibodies to angiotensine-converting enzyme of human lungs
abstract The invention relates to biotechnology and can be used in medicine and experimental biotics. The aim of the invention is to obtain a strain of hybrid cultured mouse Mus musculus cells producing monoclonal antibodies (MCAT) to the human lung angiotensin-converting enzyme (ACE). Strain A-ACE-5F1 is obtained by hybridizing immune cells of the mouse spleen BAIJB / / cg with a strain of the A-ACE-5F1 strain of the human spleen mouse BAIjB / / cg with a strain of the human A-ACE-5F1. myeloma X63-Ag8-653. It is stored in the specialized collection of transplanted somatic cells of vertebrates of the Institute of Cytology of the Academy of Sciences of the USSR under the number VSKK (P) 27D. The cells of the strain are cultivated on RPM1-1640 medium supplemented with 10% fetal bovine serum and 10% horse serum, the multiplicity of sieving is 1:10 - 1 time per 3-4 days. With the introduction of 2-5-10 cells into the abdominal cavity of BALB / C mice, ascites form after 12-16 days. MCAT production is 45-55 µg / ml culture fluid and 6-8 µg / mp ascitic. MCATs belong to the JgGj class. They specifically interact with a specific epitope of the ACE molecule and are used to detect it on tissue sections, cell membranes, biological fluids, etc. 2 tab.
