Disclosed are vectors, DNA vaccines and gene therapeutics comprising said vectors, methods for producing vectors and DNA vaccines, and gene therapeutics that contain vectors, and the therapeutic use of said vectors. In particular, vectors are disclosed which comprise (a) a nuclear anchoring protein (NAP) expression cassette comprising (i) a DNA binding domain capable of binding to a specific DNA sequence, and (ii) a functional domain capable of binding to a nuclear component, and ( b) a multimerized DNA sequence forming the binding site for the NAP anchor protein and optionally (c) one or more expression cassettes of the desired DNA sequence. Also described are vectors that do not have a papillomavirus origin of replication. NAP may be the E2 protein of bovine papillomavirus type 1 or the core antigen 1 of Epstein-Barr virus. Also described are vectors that do not have an origin of replication functional in mammalian cells and methods of expressing a desired DNA sequence in a particular subject. |
