abstract | A modified yeast strain in which acetyl-CoA pregnenolone acetyltransferase (APAT) activity is eliminated by altering the S. cerevisiae ATF2 gene or gene which encodes a protein having APAT activity and has about 60% or greater sequence identity to the YGR177C protein sequence. A transformed yeast strain in which APAT activity is eliminated by altering said gene expressing at least one enzyme from the hydrocortisone biosynthesis pathway from cholesterol selected from the cholesterol side chain cleavage enzyme (P450SCC), the 3beta-hydroxy-delta5-steroid dehydrogenase / delta5-delta4 system steroidisomerase (3beta-HSD) and 17α-steroidhydroxylase (P45017alpha). A method for in vivo oxidation of a substrate from an endogenous sterol, an exogenous sterol and an exogenous steroid using a transformed yeast strain that is cultured either alone if the strain produces an endogenous sterol, or is incubated with an exogenous sterol or an exogenous steroid and recovered the oxidized compound is optionally isolated. |
