titleOfInvention A method for separating lipoproteins from a blood gamma globulin solution
abstract Protein material containing purified human or animal gamma globulin is suspended under aseptic conditions in an amount of distilled pyrogen-free water at a temperature of 0 ° C to 6 ° C to provide a protein concentration in the range of 5 to 180 g / l. From the solution, the lipoproteins are precipitated by the addition of polyethylene glycol having a molecular weight of 4,000 to 10,000, with the resulting formulation being stirred or allowed to stand for at least 2 hours and a maximum of 24 hours at 0 ° C to 6 ° C. The lipoprotein precipitate is separated from the gamma globulin solution by centrifugation and the solution is clarified by filtration or other suitable physical method. Subsequently, the ballast compounds are separated up to a molecular weight of 150,000 in an environment of pyrogen-free distilled water or in an aqueous solution of NaCl in distilled pyrogen-free water at a concentration of up to 30 g / l at a temperature of 0 ° C to 6 ° C at pH 4.0 to 8.0 . The thus-obtained form of IgG monomer is concentrated, sterilized and stabilized by lyophilization before addition of NaCl, glucose, maltose or other sugars. The solution is filled, freeze-dried and lyophilized, and if necessary, the gamma globulin solution is subjected to further ion-exchange or affinity chromatography treatment before or after lyophilization to obtain IgG quality parameters. à
