abstract |
A polyethylene glycol derivative of a polypeptide selected from the group consisting of brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3), wherein the polypeptide is attached to polyethylene glycol is disclosed. A method of attaching a polyethylene glycol comprising one reactive aldehyde group to a polypeptide selected from the group consisting of BDNF and NT-3, wherein the polypeptide is reacted with polyethylene glycol under reductive alkylation conditions and at a pH value that is sufficiently acidic to provide the α-amino group on N the end of the polypeptide reactivity to attach the polyethylene glycol to the alpha-amino group. A homogeneous preparation of BDNF or NT-3, which are monopegylated at the α-alpha-amino group at the N-terminus for use in the treatment of impaired motoneurons and to improve brain tissue migration of BDNF or NT-3. |
