Plasmids with synthetic DNA sequences that are suitable for expression of a single first-rate rscu PA protein (i.e., the non-glycosylated portion of a single-stranded prourinokinase protein) in Enterobacteriaceae, in particular E.coli, in an amount well above that obtained from plasmids are described from the prior art. The plasmids of the invention are characterized in that their operon has one promoter that can be regulated, optionally a synthetic promoter, one Shine-Delgrano sequence, one start codon, one synthetic structural gene, and at least one terminator downstream of the structural gene. The production of plasmids as well as synthetic DNA sequences based on commercial plasmids is described. When used at high speeds, the expressed rscu-PA first-rate protein can be repackaged in a known manner to the therapeutically useful rscu Pa as a plasminogen activator, which can then be cut in the known manner to obtain a two-chain urokinase protein (rtcu-PA). and use it on a technical scale.ŕ |
