abstract | The invention provides inhibitory fusion polypeptides consisting of a TIR domain and a dimerization domain for the inhibition of TLR / IL-1R signaling pathways consisting of a TIR domain and a dimerization domain, wherein the two domains are associated with the binding peptide and wherein the polypeptide optionally also comprises a segment for membrane localization. Addition of the dimerization domain to the TIR domain significantly improves the inhibitory potential of the TIR domain. The invention also provides the use of a nucleic acid encoding a fusion-like polypeptide consisting of a TIR domain and a dimerization domain, or the use of a recombinantly produced fusion polypeptide consisting of a TIR domain and a dimerization domain, the particular use of a polypeptide additionally comprising a protein transduction domain for inhibiting cellular signaling TLR / IL-1R or for the preparation of a medicament for the treatment of chronic or acute inflammatory conditions such as infectious and autoimmune diseases, sepsis, septic shock, cancer, and others resulting from inadequate and / or signaling TLR / IL-1R. |
