abstract A method for isolating and purifying a product peptide or protein originating from cultivation of recombinant cells producing such peptide or protein using an aqueous two-phase system with an upper phase and a lower phase, comprising the steps: a) providing recombinant cells capable of producing a modified product peptide or protein containing tryptophan entities in addition to any ones naturally occuring in said product peptide or protein; b) cultivating the cells resulting from step a) under conditions resulting in expression of said peptide or protein; c) transferring the entire culture, the growth medium or harvested and disrupted cells into an aqueous two-phase system; d) allowing said system to reach an equilibrium resulting in an enrichment of said modified peptide or protein in said upper phase at an increased partition coefficient higher than that obtained with an unmodified peptide or protein; and e) recovering said modified peptide or protein from said upper phase; and peptides and proteins obtained by such method.
