abstract | Immobilisation of anchorage-dependent cells is effected by adsorption on a microcarrier (I) enzymatically degradable without significant destruction of cell surfaces. - Animal cells capable of cell division are immobilised by being entrapped in an alginate or agarose carrier are also claimed. - The (I) is suitably a naturally occurring protein or polysaccharide or deriv., and is water insoluble at ambient temp. Typically (I) is gelatin or chitosan. The (I) may be modified with a cross-linking agent, e.g. glutaraldehyde, to impart temp. resistance and increased mechanical strength. The (I) may contain particles of a magnetic material such as Fe3O4. The (I) is pref. degradable with dispase or collagenase. - The (I) is formed into particles by various treatments, e.g. by dispersing heated aq. gelatin in toluene-CHCl3 contg. surfactant. The cells immobilised in alginate or agarose are pref. beta-cells of islets of Langerhans and the prod. is suitable for the prodn. of insulin. - The immobilised cells are obtd. economically for use in cultivation processes etc. The cells can be readily removed from (I) if desired. The cells in an alginate or agarose carrier are similarly useful, e.g. in the prepn.of insulin etc. |
