abstract Bile acids, e.g. cholic acid, sulfolithocholic acid, or conjugates thereof in unextracted serum samples are assayed by: (a) providing an antiserum specific to the acid being determined, (b) mixing the unextracted serum sample with the antiserum and the same bile acid tagged with a labelling agent, (c) incubating the mixture from (b) to allow the bile acid and labelled bile acid to bind with antibodies in the antiserum, (d) separating the antibody-bound reactants from the unbound, (e) measuring the labelled bound fraction from (d) to find the quantity of bile acid in the sample by comparison with standards, wherein the assay is carried out in the presence of a binding inhibitor, e.g. 8-anilino-1-naphthalene-sulfonic acid, sodium barbital or salicylate, to inhibit retention of bile acids by binding proteins present in the serum sample.
