abstract | The invention is a new procedure for producing 9-cyclohexyl-2-alkoxy-9H-adenine derivative with the formula<IMAGE>whereby R is alkyl with 1-6 carbon atoms. The procedure is unique in that one in order1) transforms 7-amino-5-(methyltio) (1,2,5) oxadiazolo (3,4-d) pyrimidine or 7-amino-5-(methyltio) (1,2,5) thiadiazolo (3,4-d) pyrimidine with 1-2 equivalent cyclohexylamine in an inert reaction solvent at a temperature of 20-130 degrees C to produce 7-/cyclohexylamino)-5-(methyltio) (1,2,5) oxadiazolo (3,4-d)-pyrimidine or 7-/cyclohexylamino)-5-(methyltio) (1,2,5) thiadiazolo (3,4-d)-pyrimidine2) reductively formulates the obtained compounds in 50-100 percent formic acid, whereby for the oxadiazolo compound one uses catalytic hydrogenation with palladium on carbon catalysts and for the thiadiazolo compound one uses Raney nickel to produce 4-amino-6-(cyclohexylamino)-5-(formylamino)-2-(methyltio) pyrimidine3) cycles the last-named compound with alkali metal hydroxide to produce 6-amino-9-cyclohexyl-2-(methyltio) purine4) oxidizes the last-named compound in an inert solvent to produce 6-amino-9-cyclohexyl-2-(methylsulfonyl) purineand5) constrict the methylsulfone group in the last-named compound with an alkali metal alkoxide with the formula RO-alk, whereby alk is sodium or potassium and R is an alkyl group with 1-6 carbon atoms, in an inert reaction solvent to produce a 2-alkoxy-9-cyclohexyladenine with formula I. |
