titleOfInvention -Isandrographolide-19 (NAFLD) propionate for the treatment of nonalcoholic fatty liver disease
abstract The present invention relates to isoandrographolide-19-propionate for the treatment of non-alcoholic fatty liver disease (NAFLD) related to isoandrographolide-19-propionate (IAN-19P), having the structural formula : and its medicinally acceptable salts. The compound is created by reacting isoandrographolide with anhydrous propionic acid in the presence of N,N'-dicyclohexylcarbodiimide, N'-dicyclohexylcarbodiimide and 4-dimethylaminopyridine. Laboratory testing shows that isoandrographolide has lower toxicity than andrographolide, but has significantly elevated activity at 50 mM concentration. IAN-19P produces normal triglyceride levels and significantly reduces lipoperoxide formation in steatotic HepG2 cells induced with palmitate-oleate, and also reduces the leakage of alanine transaminase ALT from treated cells.
