titleOfInvention Process for preparing hyaluronidase from milt
abstract The invention relates to the production of the hyaluronidase enzyme from a promising source of raw materials - fish milk. Purpose: the possibility of using this raw material to obtain the hyaluronidase enzyme according to the previously developed technology for the preparation of the drug "Lidase" using sorption-chromatographic methods. This is achieved by the fact that fish milk is used as a raw material, crushed raw materials are subjected to acid extraction. Then the extract is centrifuged, separated. The clarified extract is filtered on a putsch filter and fed to a column of macroporous cation exchange resin. The elution of the enzyme from the carrier is carried out at pH ≅ 9.0, since at pH> 9.0 the flow of the solution is hampered by gel formation. Ammonium sulfate salting out is carried out at a degree of saturation of 0.82 - 0.85, since monomeric rather than oligomeric forms of hyaluronidase are released.
