abstract The invention relates to new sulfur-containing phosphonate compounds and new thiosubstituted compounds, including bisphosphonates, phosphonoalkylphosphonates, phosphonocarboxylates and phosphonosulfonates and their pharmaceutically acceptable salts and esters, as well as pharmaceutical compositions containing a safe and effective amount of a compound of this invention and pharmaceutically acceptable excipients, and preventing pathological conditions characterized by abnormal calcium and phosphate metabolism in humans sheep and other mammals, including the treatment or prevention of osteoporosis and arthritis, especially rheumatoid arthritis and osteoarthritis. This method consists in administering to a human or other mammal in need of such treatment a safe and effective amount of a compound or composition of the present invention or a compound and composition disclosed based thereon. The new sulfur-containing compounds of this invention have the general structural formula I described in the description. Finally, the invention relates to the treatment of arthritis in humans or other mammals in need of such treatment, comprising administering to a human or other mammal a safe and effective amount of a thiosubstituted phosphonate compound having structural formula III described in the description.
