abstract | FIELD: veterinary science; biotechnology. n SUBSTANCE: invention refers to veterinary science and biotechnology, particularly to molecular hepatology. Disclosed is a method for differential diagnosis between acute hepatitis (OH) and chronic active hepatitis (CAH), as well as early prediction of hepatic fibrosis in dogs with CAH. Expression of cfa-miRNA-122 and -21 in blood serum of dogs with primary hepatitis is analyzed. Analyses are applied in triplicate for each sample, and the obtained average cycle threshold (Ct) of each tested microRNA is normalized relative to the average ct of cfa-miRNA-16 and cel-miRNA-39-3p to obtain ΔCT. When expressing cfa-miRNA-122≥1.79 acute hepatitis is diagnosed. When expressing cfa-miRNA-122≥1.64 and cfa-miRNA-21≥1.51 expression, chronic active hepatitis and hepatic fibrosis are diagnosed in dogs. n EFFECT: invention provides creation of new effective, liver-specific non-invasive diagnostic biomarkers based on genes for early diagnosis and differentiation of OH and CAH with high sensitivity and specificity in dogs with primary hepatitis. n 1 cl, 4 dwg, 1 tbl |
