abstract | FIELD: medicine.SUBSTANCE: invention relates to laboratory diagnostics and can be used to obtain dry human blood samples and conduct further laboratory analysis for screening semiquantitative determination 11 of amino acids and/or toxic trace elements such as lead, cadmium, arsenic, thallium, mercury. For this purpose, dry human blood samples are obtained by piercing the skin cover, applying a filter card thereto with four circles applied thereon, impregnating the circle so that the blood spot is on both sides of the filter card, leaving the filter card on a flat surface face up for 4 h at room temperature avoiding sunlight. The filter card is packed in a zip bag and transported to the laboratory, where liquid chromatography is performed with tandem mass spectrometry detection using derivatising agents. Butanol in a volume of 60 mcl is therein used as a derivatising agent to determine 11 amino acids at the stage of sample preparation. The gold standard at a concentration of 1,000 ppt in 2% nitric acid is used as an extraction reagent and the Triton-X100 reagent is used as an auxiliary reagent to determine toxic substances at the stage of sample preparation.EFFECT: method ensures most accurate detection of certain substances, namely, 11 amino acids and some toxic elements, due to the use of specific derivatising and extraction reagents for each of the detected substances, wherein the procedure is significantly accelerated and made cheaper due to the proposed technique for obtaining blood samples in the form of "dry spots", available for the patient to receive independently and send to the laboratory from home.3 cl, 5 tbl, 4 ex |
