abstract | FIELD: biotechnology.SUBSTANCE: invention relates to the field of biotechnology, veterinary microbiology and genetic engineering and can be used for the diagnosis of histophilia in cattle. A set of oligonucleotides for identification and detection of Histophilus somni bacterium DNA by real-time PCR contains universal forward (Hsmn_F) and reverse (HsmnR) primers and a fluorescently labeled probe (Hsmn Z) for identification of Histophilus somni bacterium, complementary regions of 16S ribosomal RNA of the microorganism Histophilus somni, having the declared nucleotide composition. A method for identifying and detecting Histophilus somni bacterium DNA by real-time PCR includes isolating DNA from biological material, staging a polymerase chain reaction using a set of oligonucleotides for identifying and detecting Histophilus somni bacterium DNA in a reaction mixture. Real-time PCR is performed using an established amplification program. The results of the real-time amplification are evaluated using the software, and the results are interpreted based on the presence (or absence) of the intersection of the fluorescence curve with the cut-off line set at 0.05.EFFECT: effective diagnostic histophilia in cattle.2 cl, 2 ex, 1 tbl, 1 dwg |
