titleOfInvention Method for activation of trypsin
abstract FIELD: chemistry.SUBSTANCE: invention relates to a method for activation of trypsin. In implemention of the method, a 0.5 to 1.0% solution of trypsin is mixed in a 0.1 M phosphate buffer solution with a pH of 7.5 to 8.0 and a temperature of the solution of +28 to 30°C in a magnetic stirrer with heating for 15 to 20 min with a rotation frequency of the stirring bar of 500 to 1500 rpm. Ultrasonic treatment is performed for 5 to 10 minutes, the homogenised solution is treated with blue spectrum light with a wavelength of 430 to 460 nm and a light flux power of 35 mcW/cm2at a distance of 10.0 to 9.0 cm from the monochrome emitter. The activity of trypsin is determined by the cleavage of a gelatin solution solidified at a temperature of +16 to 18°C with an increased mass fraction of gelatin of 23 to 25% preliminarily swollen for 60 to 80 minutes, stirred for 30 to 45 minutes until complete dissolution of the granules of gelatin filtered through a nylon sieve, with depressions appearing in the layer of solidified gelatin as a result of applying 0.10 to 0.15 ml of 0.5 to 1.0% trypsin solution in a 0.1 M phosphate buffer solution with a pH of 7.5 to 8, 0 at a ratio of 1:2, 1:8, 1:16, 1:32, 1:64, 1:128, 1:192, 1:256 and leaving for 15 to 17 minutes and then washing the gelatin layer with a directed cold water stream at a temperature of +6 to 8°С.EFFECT: invention ensures increase of activity of trypsin.1 cl, 6 tbl, 3 ex
