abstract | FIELD: biochemistry.SUBSTANCE: group of inventions relates to a method for the production of L-fucose in free form using a recombinant microorganism and a recombinant microorganism for the production of L-fucose in free form. The method includes culturing the recombinant microorganism in a medium suitable for the growth of the microorganism, where the microorganism is unable to metabolize L-fucose, so that L-fucose accumulates during the cultivation step, and isolating free L-fucose from the medium. The recombinant microorganism for the synthesis of L-fucose contains a heterologous enzyme that catalyzes the hydrolysis of GDP-L-fucose for the release of L-fucose from GDP-L-fucose, wherein the above-mentioned enzyme is a variant of alpha-1,2-fucosyltransferase encoded by the wbgL gene from E. coli: O126, containing two amino acid substitutions selected from the group consisting of (i) substitutions of histidine 124 for alanine and glutamate 215 for glycine, and (ii) substitutions for asparagine 69 for serine and isoleucine 268 for proline.EFFECT: group of inventions provides efficient production of target monosaccharide.9 cl, 4 dwg, 1 tbl |
