titleOfInvention | Mutant microorganisms resistant to death under the action of lactose |
abstract | FIELD: microbiology.SUBSTANCE: invention relates to a method for producing microorganisms resistant to the phenomenon of suppression by the action of lactose, to microorganisms obtained by using this method. A method for producing microorganisms resistant to the phenomenon of lactose suppression, grown in an environment in which lactose is combined with another carbon source, involves changing the expression of the lactose transporter in microorganisms by introducing a heterologous promoter in front of an endogenous or exogenous lactose transporter gene or by changing the codons of the endogenous lactose transporter gene to codons used by the microorganism more often, or to codons used by the microorganism less often, the subsequent cultivation of these microorganisms on a medium that includes a carbon source other than lactose, as well as the addition of lactose to the specified medium, including a carbon source other than lactose, during the growth of these microorganisms and the detection of the lactose transporter expression. Selection of microorganisms resistant to the phenomenon of suppression by the action of lactose, growing on the specified medium, including lactose and another carbon source, which retain at least 50% of the level of lactose transport into the cell, achieved with the wild-type expression cassette of the specified lactose transporter, is carried out on based on the detected expression of the lactose transporter. At the same time the specified carbon source other than lactose is selected from the group consisting of glycerin, maltose, glucose, fructose, sucrose, fucose, mannose, sialic acid, starch, cellulose, polyols, organic acids and pentoses. Also proposed are variants of microorganisms obtained by this method. An expression cassette of a lactose transporter is also proposed, leading to the expression of a lactose transporter, which does not lead to the phenotype of suppression by the action of lactose, which is isolated from a variant of the specified microorganism. he specified expression cassette and the specified variants of microorganisms are used for the production of 2-fucosyllactose, 2'-fucosyllactose, 3-fucosyllactose, 2', 3-difucosyllactose, lacto-N-triose, lacto-N-tetraose, lacto-nN-tetraose, 3'-sialyllactose, 6'-sialyllactose, glycolipids or galactosylated compounds. A method for producing an expression cassette is also proposed.EFFECT: invention provides for the preservation in the resulting microorganism of at least 50% of the level of lactose transport into the cell, achieved with the wild-type expression cassette of the specified lactose transporter, based on the detected expression of the lactose transporter.14 cl, 1 tbl, 21ex, 21 dwg |
