titleOfInvention | Method for sensibilization of plate for enzyme immunoassay by insoluble protein antigens |
abstract | FIELD: medicine; pharmaceuticals.SUBSTANCE: invention relates to medicine and pharmaceutics, namely to a method for sensibilization of a plate for enzyme immunoassay by insoluble protein antigens of a plague microbe, involving destruction of bacterial mass by ultrasound followed by separation of subfractions, affinity chromatography of proteins, separation and dissolution of purified target protein precipitate with denaturation of urea and determination of their concentration in solutions, characterized by that a solution of denatured in purified urea fibres MetQ, OmpA, Fba and YapM is added to the wells of the plate and subsequent ten-fold dilution in the wells with a carbonate-bicarbonate buffer solution, accompanied by renaturation of proteins and their passive immobilisation in polystyrene plates.EFFECT: technical effect consists in sensitizing polystyrene plates with protein antigens, insoluble in standard buffer systems used at the first stage of treatment, and performing ELISA for such protein antigens, thereby increasing sensitivity and specificity of the test.1 cl, 5 dwg, 1 tbl, 5 ex |
