FIELD: medicine.SUBSTANCE: present invention refers to medicine, namely to a method for preparing the agent for cell-mediated gene therapy. Method for preparing the agent for cell-mediated gene therapy of whole blood with successive production of whole blood the leuko-platelet layer and leuco-concentrate from leuko-platelet layer, in which hydroxyethyl starch is added at ratio of 1:1, the obtained mixture of leuko-platelet layer with hydroxyethyl starch is centrifuged with subsequent production of supernatant containing plasma, hydroxyethyl starch and leukocytes, then physiologic saline is added to the supernatant, centrifuged to produce a leuco-concentrate, in which a recombinant replicative-defective viral vector Ad5/35F is added, created on the basis of human adenovirus 5 serotype with modified fiber and bearing a recombinant gene of reporter green fluorescent protein in ratio of leukocyte : viral vector – 1:5, delivering recombinant genetic material in CD46 – positive leukocytes.
