FIELD: biotechnology. n SUBSTANCE: invention relates to biotechnology and molecular biology. Disclosed are methods and compositions for modifying one or more target loci in a cell. Such methods include providing a cell containing a first polynucleotide coding a first selective marker operably linked to a first promoter active in a cell, wherein the first polynucleotide further comprises a first recognition site for the first nuclease agent. First nuclease agent is introduced into the cell, wherein the first nuclease agent induces a single- or double-strand break at the first recognition site. First targeting vector containing a first polynucleotide insert flanked by the first and second homologous arms, which correspond to the first and second target sites located close enough to the first recognition site, is additionally introduced into the cell.
