titleOfInvention | Ureohydrolases as dominant selective markers in yeast |
abstract | FIELD: chemistry.SUBSTANCE: invention relates to molecular biology. Disclosed is a method of culturing a microorganism selected from genera Saccharomyces sensu stricto, Kazachstania, Naumovozyma, Nakaseomyces and Vanderwaltozyma, in the presence of guanidinobutyrate as the only nitrogen source, including the incorporation into the said microorganism of a nucleic acid molecule containing a nucleotide sequence coding guanidinobutyrase with number EC 3.5.3.7 and operably linked to promoter and terminator sequences, culturing said microorganism such that a nucleic acid molecule encoding guanidinobuturase is expressed in said microorganism, and culturing said microorganism in the presence of guanidinobutyrate as a single nitrogen source. Also disclosed is a microorganism for growth on guanidinobutyrate as a single source of nitrogen, selected from genera Saccharomyces sensu stricto, Kazachstania, Naumovozyma, Nakaseomyces and Vanderwaltozyma containing nucleotide sequence coding guanidinobutirase with EC number 3.5.3.7. Also disclosed is a method for changing the genome of a microorganism specified in genera Saccharomyces sensu stricto, Kazachstania, Naumovozyma, Nakaseomyces and Vanderwaltozyma.EFFECT: invention provides efficient use of guanidinobutyrase as a selective marker.10 cl, 10 dwg, 5 tbl, 1 ex |
