abstract | FIELD: biotechnology; medicine. n SUBSTANCE: group of inventions relates to biotechnology and medicine, in particular to modified glycoprotein hormones having superagonist activity, and the use thereof in the treatment of conditions associated with glycoprotein hormone activity. Proposed is a modified alpha subunit polypeptide selected from a group consisting of a wild type polypeptide of the alpha subunit of bovine (SEQ ID NO: 2), a wild type polypeptide of the alpha subunit of sheep (SEQ ID NO: 3), a wild type polypeptide of the alpha subunit of horse (SEQ ID NO: 4), a wild type polypeptide of the alpha subunit of pig (SEQ ID NO: 5), with at least one basic amino acid substitution of the wild-type amino acid at position 15, 17, 18, 20 and/or 24 and one, two or three amino acid inserts introduced immediately after position 6 or 7. Said modified alpha subunit polypeptide is capable of binding to a beta subunit polypeptide to form a hormone of glycoprotein hormones. Formed modified glycoprotein hormone is used to stimulate ovulation. n EFFECT: resultant analogues of long-acting glycoprotein hormones demonstrate enhanced bioactivity both in vitro and in vivo as compared to wild type counterparts, they are useful for treating subjects showing low receptor expression or poor receptor responsiveness, and for the treatment of any condition associated with glycoprotein hormone activity. n 9 cl, 11 dwg, 8 tbl |
