titleOfInvention | Method for enzymatic production of l-methionine from o-phospho-l-homoserine and methanethiol |
abstract | FIELD: chemistry. n SUBSTANCE: method for the preparation of L-methionine in which O-phospho-L-homoserine (OPHS) and methanethiol are enzymatically converted to L-methionine and H 3 PO 4 according to the reaction scheme: O-phospho-L-homoserine + CH 3 -SH <=> L-methionine + H 3 PO 4. This enzymatic conversion is achieved by using OPHS-dependent methionine synthase, which is a protein obtained from cystathionine-gamma synthase (CF 2.5.1.48) as a result of a mutation. OPHS-dependent methionine synthase is a protein selected from the group consisting of (a) a protein comprising any of the amino acid sequences SEQ ID NO: 6 to 29; (b) a protein having a sequence identical to at least 90% of any one of SEQ ID NOs: 6 to 29, and having enzymatic activity of converting OPHS and methanethiol to L-methionine and H 3 PO 4 . Also disclosed is the use a protein having enzymatic activity of converting OPHS and methanethiol to L-methionine and H 3 PO 4 , for converting OPHS and methanethiol to L-methionine and H 3 PO 4 . Use of the method of producing L-methionine enables to produce S-adenosyl methionine, cysteine, glutathione, as well as 2-oxo-4-methylthiobutanoate. n EFFECT: group of inventions enables to efficiently convert OPHS + CH 7 -SH into L-methionine. n 8 cl, 7 dwg, 3 tbl, 6 ex |
