FIELD: biochemistry. n SUBSTANCE: invention relates to biochemistry. Proposed is a method of a multi-specific binding molecule preparation comprising at least two binding moieties, such as an antibody, where the method comprises culturing a cell containing a nucleic acid encoding soluble A sortase from S. aureus with a C-terminal retention signal in the endoplasmic reticulum, nucleic acid encoding a first binding group comprising a sortase motif at its C-end, followed by a retention signal in the endoplasmic reticulum, and a nucleic acid encoding a second binding group containing at least one diglycine at its N-end.
