titleOfInvention Method for molecular-genetic intraspecific typing of toxygenic vibrio cholerae o1 eltor strains
abstract FIELD: biotechnology. n SUBSTANCE: method for molecular-genetic intraspecific typing of toxigenic Vibrio cholerae O1 Eltor strains is claimed. The method involves isolation of DNA from the test Vibrio cholerae O1 Eltor strain, PCR conduction with the specific primers, and taking into account the results obtained. Amplification of the test DNA is performed by the designed primers to each of the ICE elements of Indian and Mozambican types, in the presence of intercalating Siber Green dye. The amplification products are then subjected to melting, the results are taken into account by temperature melting peaks and the amplicon size. At the melting point of 87°C and the amplicon size of 300 bp, presence of ICE of Indian type is detected. At the melting point of 79°C and the amplicon size of 149 bp, presence of ICE of Mozambican type is detected. n EFFECT: invention allows to quickly and reliably determine the origin of isolated toxigenic of V cholerae O1 Eltor strains. n 1 dwg, 4 tbl, 3 ex
