abstract | FIELD: biotechnology. n SUBSTANCE: invention relates to biotechnology. Invention describes a method of identifying pairs of DNA or RNA fragments, initially present in the same living or fixed cells, in particular method of identifying native pairs of genes of light and heavy chains of antibodies, as well as native pairs of alpha- and beta-chains of T-cell receptor (TCR). Method is based on emulsion PCR or RT-PCR, carried out on living or fixed cells, during which PCR-amplification of DNA or RNA fragments pairs takes place, followed by physical coupling of PCR amplification products. All the above reactions are carried out within emulsion with separate cells, each of which is placed in separate emulsion drop. Method is characterized by new type of PCR-lock, which enables, selective amplification of library of gene fragments coupled in emulsion after emulsion reactions. Selective suppression of PCR participation of uncoupled fragments enables amplification mainly of target pairs of fragments, previously successfully coupled within drops in emulsion compared to amplification of pairs of fragments, capable of non-target coupling after extraction of nucleic acids' mixture from the emulsion. n EFFECT: method of identifying pairs of DNA or RNA fragments, initially present in the same living or fixed cells is proposed. n 9 cl, 7 dwg, 5 tbl, 1 ex |
