abstract | FIELD: chemistry. n SUBSTANCE: invention relates to analytical chemistry and specifically to methods of standardisation of medicinal preparations, medicinal plant raw materials, products of food, chemical and cosmetic industry by content of glutamic acid, and can be used in pharmaceutical, chemical and cosmetic industry. Method of identifying and determining degree of purity and quantitative content of glutamic acid using high-performance thin-layer chromatography consists in dissolving of analysed preparation batch in water with subsequent using silica gel plates of “Sorbfil" brand with aluminium or polymer substrate PTSH-P-A or PTSH-AF-A. Eluent used is system composed of n-butanol-glacial acetic acid-water (4:1:1), and 1 % ninhydrin alcohol solution as developer. Optimal sample volume is 1 mql of aqueous solution with glutamic acid content of 1 mg/ml. At that, chamber saturation time with eluent vapors is 40 minutes, elution time is 55 minutes, plates holding time in thermostat after development at t° = 103±2 °C is 3-5 minutes. After development of chromatographic zones, plates are scanned by means of flatbed scanner, and obtained images are processed using known “Sorbfil Videodensitometer” software, glutamic acid zone is identified at specific value R f in comparison with reliable standard sample, and quantitative content of glutamic acid (c, mqg) in sample applied on chromatogram, is calculated by formula wherein S is area of chromatographic zone on chromatogram, calculated using “Sorbfil Videodensitometer” software. n EFFECT: technical result is high accuracy, as well as possibility of simultaneous authentication, degree of purity and quantitative determination of glutamic acid in medicinal preparations, medicinal plant raw material, articles of food, chemical and cosmetological industries, as well as possibility of separation of complex mixtures and separation of glutamic acid from other biologically active substances. n 1 cl, 8 dwg, 3 tbl |
