titleOfInvention | Separation of non-soluble target proteins |
abstract | FIELD: chemistry. n SUBSTANCE: claimed invention relates to biotechnology. Claimed is method for separation of recombinant non-soluble target silk protein from suspension of intact or destroyed host cells. Claimed method includes obtaining suspension of intact or destroyed host cells, which contain recombinant non-soluble silk protein, addition of base water solution to said suspension to destroy host cells and for solubilisation of non-soluble parts of host cells, in which silk protein remains unsolved in suspension, separation of recombinant non-soluble silk protein from solubilised parts of host cells. Base represents metal or ammonium hydroxyl, and base water solution has pH values from 9 to 14. n EFFECT: claimed separation method makes it possible to obtain recombinant silk protein with high output and purity due to effective separation of non-soluble recombinant silk protein from solubilised non-soluble parts of host cells, with claimed method being fast and easily realised purification method. n 22 cl, 1 tbl, 4 ex, 6 dwg |
