FIELD: chemistry. n SUBSTANCE: invention relates to biotechnology and specifically to methods of detecting a target nucleic acid sequence in DNA or a nucleic acid mixture by PTOCE (PTO cleavage and extension) assay. The method includes detecting a target nucleic acid sequence, wherein PTO (probing and tagging oligonucleotide) hybridised with the target nucleic acid sequence is cleaved to release a fragment. The fragment is hybridised with the CTO (capturing and templating oligonucleotide) to form an extended duplex, followed by detecting the presence of the extended duplex. The extended duplex provides signals from labels indicating the presence of the extended duplex and has an adjustable T “пл” value, which is well suited for detecting the presence of the target nucleic acid sequence. n EFFECT: invention enables to detect a target nucleic acid sequence in DNA or a nucleic acid mixture with high efficiency. n 57 cl, 30 dwg, 7 ex |
