abstract FIELD: chemistry. n SUBSTANCE: invention relates to biotechnology and represents a method of obtaining a polypeptide in a culture of mammalian cells (versions) and a medium for cultivation of the mammalian cells. The claimed invention can be used for obtaining the polypeptide of interest in great amounts. The method includes obtaining the cell culture, which contains the mammalian cells containing a nucleic acid, which codes the polypeptide of interest, and an initial medium for cell cultivation, with a volume of the initial medium for cell cultivation constituting approximately 60-99% of a volume of the desired cell cultivation medium. The nutritional medium for cultivation is added to the cell culture, with a volume of the nutritional medium for cell cultivation constituting approximately 1-40% of a volume of the cell cultivation medium. The obtained medium for cell cultivation represents a medium for cell cultivation, containing from approximately 7 mM to approximately 30 mM of leucine, from approximately 7 mM to approximately 30 mM of lysine, from approximately 7 mM to approximately 30 mM of threonine, from approximately 7 mM to approximately 30 mM of proline and from approximately 7 mM to approximately 30 mM of valine.
