abstract | FIELD: medicine, pharmaceutics. n SUBSTANCE: group of inventions relates to methods of identifying allergenic proteins and peptides. In particular, the claimed invention relates to methods of identifying allergenic proteins and/or peptides of milk: alphaS1-, alphaS2-, beta- or kappa-casein, including the following stages: obtaining at least one expression library, containing DNA or cDNA, obtained from tissue of a mammary gland of a lactating mammal, preferably, a lactating cow, expression of at least one protein or peptide, coded by the said expression library; determination of binding ability of at least one protein or peptide with IgE of at least one serum of an individual, sensitive to mammalian milk, preferably to cow's milk; contact of at least one protein or peptide, demonstrating IgE-binding ability, determined at a stage c) with basophilic cells, eosinophilic cells or mast cells, and identification of at least one protein or peptide as an allergen, if the said basophilic cells, eosinophilic cells or mast cells release at least one mediator after the contact with the said at least one protein or peptide of a stage d). n EFFECT: claimed methods are highly efficient in determination of allergenic proteins and peptides in various biological sources. n 13 cl, 13 dwg, 3 tbl, 4 ex |
