titleOfInvention Method of obtaining tracheobronchial bioimplant
abstract FIELD: medicine. n SUBSTANCE: invention relates to medicine and veterinary, in particular, to transplantology. In order to obtain bioimplant mechanical cleaning of donor's tissue of trachea/bronchi from connective and adipose tissues is carried out. After that donor's tissues of trachea/bronchi are washed with distilled water and incubated for 10-14 days in 5-10% solution of chlorine-containing oxidant. After that tissues of trachea/bronchi are washed with physiological solution and cryoconservation (in case of necessity) is performed. Mesenchymal stem cells CD271+ are isolated from recipient's bone marrow by method of immunomagnetic separation and their generation is carried out. Suspension of recipient's epithelial cells is obtained. Matrix of trachea/bronchi is covered with protein of recipient's blood serum and surface of bioimplant is inoculated with epithelial and mesenchymal cells. Bioimplant is incubated in RPMI-1640 medium until implantation. n EFFECT: method ensures short cycle of preparation (15-21 days) of individualised bioimplant based on sterile matrix with preservation of framework function of organ, covered with protein-cellular envelope, which do not possess heterogenic immune properties. n 25 dwg, 4 ex
