FIELD: chemistry. n SUBSTANCE: invention relates to biochemistry. Disclosed are methods of extracting a purified protein product (versions) and methods of extracting a purified antibody (versions) via chromatography in weak distribution mode. The methods involve passing the loaded liquid containing a protein product or antibody through a medium at operating conditions which, in one version of the method, facilitate binding of at least 1 mg of product per 1 ml of the medium. In another version of the method, operating conditions are characterised by a distribution factor equal to at least 0.1. Said operating conditions are determined by screening. Said medium is selected from a group comprising a charged ion-exchange medium, a hydrophobic interaction chromatography resin and an immobilised metal-affinity chromatography resin. The purified product is then separated from the eluate coming from the column. n EFFECT: methods ensure high output of the purified protein product or antibody while using high loads of 50-500 mg of product per ml of medium. n 184 cl, 15 dwg, 30 tbl, 8 ex |
