titleOfInvention | METHOD OF FLUORESCENCE IMMUNOASSAY OF OESTROGEN β RECEPTORS IN HUMAN SOLID TUMOURS |
abstract | FIELD: medicine. ^ SUBSTANCE: for oestrogen (OR) receptor analysis, single-cell suspensions of a tumour tissue and a reference culture which is a monolayer culture of MCF-7 line human breast cancer are prepared in solid tumours. The cells are fixed in 4 % formaldehyde, incubates with Tween 20, washed by means of phosphate buffer pH 7.4. Then they are incubated with primary and secondary antibodies, twice washed by means of phosphate buffer pH 7.4, and fluorescence is measured in a flow cytofluorimetre. The data are analysed by calculating the D value of a Kolmogorov-Smirnova statistical test. A percentage of specifically coloured cells is the D value multiplied by 100. ^ EFFECT: invention enables differential and strictly quantitative evaluation of OR expression in tumour tissue. ^ 2 dwg |
