abstract | FIELD: veterinary. n SUBSTANCE: method of carrying out express NBT-test of leukocytes in peripheral blood of dogs includes mixing of blood with dissolved nitro blue tetrazolium (NBT), incubation, microscoping and calculation of positive and negative cells. In test-tube introduced and mixed are: 0.05 ml of fresh heparinised blood, 0.05 ml of 0.2% NBT solution on phosphate-salt buffer, then said mixture is incubated during 30 minutes at T=37 degrees Celcius, after the end of incubation 0.3 ml of 3.5% acetic acid solution is introduced into test tube, thoroughly mixed; aliquot of obtained mixture is taken and used to charge Goryaev chamber, cells are microscoped under magnification 40×10, ensuring minimal visibility of non-stained cells, NBT-positive cells, which contain formazan in blood cytoplasm, and NBT-negative cells, which do not contain formazan, are calculated in differentiated way. n EFFECT: creation of simple, fast and efficient method of carrying express NBT-test in leukocytes of peripheral blood of dogs. |
