abstract | FIELD: technological processes; veterinary medicine. n SUBSTANCE: on the basis of genomic DNA of birds adenovirus CELO two plasmids are designed, one of which contains its left fragment (1-20028 bps), which includes expressing cassette under control of own main late promoter of adenovirus MLP, and the other contains the right fragment of genome CELO (17,390-43,804 bps) with zone that is homologous to nucleotide sequence of the first plasmid for performance of recombination, and also with deletion in 3,787 bps for increase of packing capacity of adenovirus CELO. Expressing cassette is cloned in the area of deletion, which consists of exogenic promoter that provides high level of expression in eukaryotic cells, polylinkers for cloning of one or more target genes and signal of polyadenylation. Homologous recombination between designed plasmids is performed in the culture of cells of leghorn rooster hepatoma (LMH), which provides preparation of recombinant birds adenovirus CELO. For cloning in expressing cassettes genes of cytokines, growth factors, antigens and oncosuppressors of human being, birds and microorganisms are used, both taken separately and in certain combinations for achievement of required genetic therapeutic effect or high level of immunisation. n EFFECT: creation of genetically engineered recombinant vaccines of new generation and preparations for genetic therapy. n 32 cl, 4 dwg, 22 ex |
