abstract | FIELD: biotechnology, medicine, vaccines. n SUBSTANCE: invention relates to a method for preparing live vaccine for prophylaxis of influenza. Method involves infection of chicken embryos with the vaccine influenza strain, collection of virus-containing allantois fluid, addition of a stabilizing agent, sterilizing filtration, pouring into ampoules and lyophilization. Stabilizing agent comprises specimens of the following components, g: sucrose, 135; lactose, 90; glycine, 30; sodium glutamate, 15; tris-buffer, 5 and sodium chloride, 7.5, dissolved in 0.7 l of sterile distilled water and mixed with 10% gelatin solution in the ratio = 3:1. In the lyophilization process ampoules with vaccine are kept at temperature -35°C for 5 h being for the first 2 h ampoules are kept under atmosphere pressure (50 ± 10) mm of mercury column and then at temperature -16°C for 30 h. Then a temperature regulator is switched off for 6 h followed by additional drying at temperature +28°C for 15 h, the following filling ampoules with argon and sealing. Prepared vaccine comprises reduced amount of protein that decreases the possibility for arising allergic response reactions.
