FIELD: biochemistry, gene engineering, in particular amidation of hormones and other peptides used in medicine and agriculture. n SUBSTANCE: peptide sequence obtained in process of triptic digesting of peptidylglycine alpha-amidating monooxygenase (PAM) from fat thyroid encephaloid carcinoma is determined. On the base of obtained data oligonucleotide sequences are synthesized and used for isolation of full-sized PAM encoding DNA. Expression of obtained DNA fragments in heterogeneous systems makes it possible to obtain recombinant PAM useful in amidation reaction of peptide substrate with C-terminated glycine providing the same effect that for native enzyme. n EFFECT: effective method for production of amidated peptides. n 12 dwg, 2 tbl, 9 ex |
