abstract FIELD: molecular biology, genetic engineering, medicine, pharmacy. n SUBSTANCE: invention relates, in particular, to a method for identifying genes taking part in adaptation of body to the environment of its residence. Invention proposes a method for identifying a microorganism with reduced adaptation to the specific environment. Method involves preparing multiple microorganisms each of that is mutated independently by insertion inactivation of nucleic acid gene comprising the unique marker sequence so each mutant comprises different marker sequences, or clones of indicated microorganism, stage (1). Individual storage of each mutant sample prepared at the stage (1) and individually accumulated nucleic acid comprising the unique marker sequence from each individual mutant, stage (2). Addition of multiple mutants at the stage (1) to indicated specific environment and providing to these microorganisms that able to this the possibility to growth in indicated environment medium, stage (3). Isolation of microorganisms from indicated environment or its selected part and isolation of nucleic acid from these microorganisms, stage (4). Comparison of marker sequences in nucleic acid isolated at the stage (4) with the unique marker sequence of each individual mutant accumulated at the stage (2), stage (5). Selection of the individual mutant that doesn't comprise any of marker sequences isolated at the stage (4), stage (6). Also, invention proposes DNA (variants) responsible for reducing adaptation of microorganism to environment isolated from genome Salmonella, the virulence gene comprising this DNA, and polypeptide encoded by this gene. Except for, invention proposes a method for assay of compound reducing adaptation of microorganism to environment. The proposed method involves selection of such compound that inhibits the function of gene comprising DNA responsible for reducing adaptation of microorganism to environment. Also, invention proposes an anti-sense nucleic acid obtained by above indicated method.
