abstract FIELD: organic chemistry, chemical technology, medicine, pharmacy. n SUBSTANCE: invention relates to a method for preparing calcium folinate. This product belongs to the most important and vital essential medicinal agents and shows the broad field in applying: as antidote of methotrexate and other anti-metabolites used in chemotherapy of oncological diseases, as an effective formulation of vitamin folic acid used in treatment of anemia, among them, post-irradiation ones and also for compensation of folate insufficiency in deficiency in folate reductase activity in body, and so on. Method involves stages for preparing an intermediate substance, i. e. 5,10-N,N-methenyltetrahydrofolate by photochemical reduction of folic acid by effect of long-waved ultraviolet radiation (UV-light) in medium of inert gas and inert solvent, such as isopropyl or methyl alcohol, or ionized water in the presence of electron donor, such as ascorbic or glycolic acid in combination with the coupled formulation with formic acid. Calcium folinate is prepared by conversion of 5,10-N,N-methenyltetrahydrofolic acid to chloride form and the following hydrolysis in the presence of amines. Hydrolysis results to opening the imidazoline ring to form 5-formyltetrahydrofolic acid that is precipitate as calcium salt, i. e. calcium folinate.
