abstract | FIELD: biotechnology, molecular biology, genetics. n SUBSTANCE: invention can be used for in vitro analysis in transcription of viral and cellular genes. Cell-free transcription of DNA matrix in vitro is carried out using enriched extract from cellular nuclei and marked nucleotide and marked transcript is determined. DNA matrix comprises unique restriction sites for site-specific restriction endonucleases: Pst I, Eco RI, Sac I, Kpu I, Sac II, Bam HI, Swa I, part of plasmid pUC19, five sites for binding yeast protein Ga14, "TATA"-block of human receptor of T cells Vβ 8.1 between restriction sites Sac II and Bam HI, initiating site of adenovirus of the main later promoter between restriction sites Bam HI and Swa I and G-free sequence with length about 800 nucleotides.
